ตัวเกม Aliens: Dark Descent จะให้ผู้เล่นสวมบทบาทเป็นผู้บังคับบัญชาที่ต้องคอยสั่งการหน่วยทหารเดนตาย Colonial Marines เข้าไปทำภารกิจหยุดยั้งระงับเหตุแพร่ระบาดของอสูรซีโนมอร์ฟบนดวงจันทร์ Moon Lethe โดยผู้เล่นต้องนำทีมทหารเข้าต่อสู้กับกองทัพฝูงเอเลี่ยนหลากหลายประเภททั้ง Facehuggers, Praetorians, Alien Queens รวมไปถึงศัตรูจากฝ่าย Weyland-Yutani Corporation ที่เป็นมนุษย์ด้วยกัน แถมยังมีศัตรูหน้าใหม่ที่จะปรากฏตัวให้เห็นเป็นครั้งแรกเฉพาะในเกมนี้เท่านั้น

ผู้เล่นจะได้บุกแทรกซึมด่านเลเวลที่มีลักษณะเปิดกว้างและเลือกสั่งการยูนิตของคุณเข้าจัดการกับศัตรูผ่านระบบควบคุมที่สะดวกง่ายดายไม่ว่าจะกดปุ่มบนจอยหรือใช้เมาส์คลิก งานหลักของเราคือการทำทุกวิถีทางเพื่อให้สมาชิกหน่วยทุกคนยังมีชีวิตรอด อาทิเช่น เลือกเส้นทางที่ดูปลอดภัย, มองหาเส้นทางลัดชอร์ตคัต, สร้างพื้นที่เซฟโซน ไปจนถึงติดตั้งอุปกรณ์เตือนภัยตรวจจับการเคลื่อนไหว เนื่องด้วยในเกมนี้ถ้าหากตัวละครของคุณตัวใดเกิดตายขึ้นมา เขาคนนั้นจะ ตายถาวร หายไปจากเกมมิอาจฟื้นหวนคืนกลับมาได้อีก

สำหรับคลาสตัวละครหน่วยทหาร Marines เริ่มต้นจะมีอยู่จำนวน 5 คลาสอาชีพให้คุณได้ใช้งาน แต่ละคลาสล้วนมีอบิลิตี้ความสามารถและอาวุธถนัดที่โดดเด่นเฉพาะแตกต่างกันออกไป ซึ่งคุณจะได้เลเวลอัพปรับแต่งพวกเขาไล่ตั้งแต่อาวุธ ชุดเกราะ ยันสกิลเพิร์ค เพื่อให้พวกเขาพร้อมเผชิญรับมือภารกิจมิชชั่นที่ยากท้าทายและเสี่ยงตายขึ้นเรื่อยๆ
